One fourth of the human cerebral cortex and nearly twenty percent of all brain activity is devoted to sight; we are well adapted to analyse images and see small changes in two-dimensional rather than one-dimensional signals.

An example of our proteome imaging focus is the collaboration we have had with the SIB over the last several years in developing algorithms that have been incorporated into the Melanie software package.

Since 1984, the SIB has been developing the Melanie algorithm and imaging platform for 2-D gel analysis. The software is recognized as a leader in the field of two-dimensional gel electrophoresis (2-DE). Distributed by GeneBio since 1999, it also powers GE Healthcare's ImageMasterTM 2D Platinum platform.

melanie The most recent product releases are Melanie 7.0 and Melanie 7.0 DIGE. These versions provide powerful and innovative solutions to automate and shorten the path from data acquisition to protein information.

The superior spot detection and matching algorithms facilitate the extraction of statistically valid differences between groups of 2-D gels, while requiring minimal user intervention and therefore speeding up analysis time. The application integrates filtering, querying, reporting, statistical and graphing options so that you can easily view, compare, analyse and present your results. Melanie 7.0 DIGE extends the benefits of this powerful analytical software to 2-D DIGE experiments, eliminating gel-to-gel variation and improving accuracy by enabling the use of an internal standard.

New Applications For Melanie
MSight Taking advantage of the SIB's twenty year track record in proteome image analysis, Melanie has recently been used for a new imaging tool, MSight, devoted to mass spectrometry. As most current proteomics experiments are aimed at the quantitative profiling of proteins in complex mixtures using high-resolution separations followed by mass spectrometry, MSight was specifically developed for the representation of mass spectra along with data from the separation step, such as liquid chromatography (LC). The software allows graphical exlploration inside huge datasets, as well as image and analysis tools, thus giving scientists access to informationn that was previously hidden.
  • Fast multi-resolution visualisation of the data as an image ensuring seamless transition from a global overview of all spectra to selected isotopic peaks
  • User-friendly navigation through large volumes of data
  • Visualization tools to discriminate peptide or protein from noise
  • Differential proteome analysis by comparing several sets of data, providing fast and intuitive detection of significant qualitative and quantitative differences
  • Assessment of the data quality and the experimental design is simplified by providing a direct means to verify key aspects, such as the quality of the separation, the presence of artefacts and contaminants or mass calibration problems
  • Reads MS and MS/MS files generated by the majority of mass spectrometers
